马氏钳蝎蝎毒对艾氏腹水癌小鼠和Hela细胞株的作用

摘    要:应用马氏钳蝎蝎毒(Scorpion venom cruds,SVC)腹腔注射于艾氏腹水癌(Ehrlich ascitescarcinoma,EAC)小鼠以及处理体外培养的人宫颈癌(Hela)细胞株,观察SVC在体内、体外的抗肿瘤作用。结果显示,SVC 0.1mg/kg,0.3 mg/kg腹腔注射10 d,EAC小鼠的生命延长率分别为52.04%、54.38%.差异显著(P<0.05);在停药第10 d时,EAC小鼠的体重抑制率分别为31.1%、24.2%,亦有明显差异。实验重复两次,均表明SVC对小鼠的生存期有明显延长作用以及对小鼠体重的增长有抑制作用。MTT检测法显示,SVC体外处理EAC细胞,在一定浓度范围内对EAC细胞有明显的细胞毒性作用。SVC对体外培养的Hela细胞作用不明显。提示SVC可能具有一定的抗癌谱。

Abstract:The mice with Ehrlich ascites carcinoma (EAC) was injected with Scorpion venom crude (SVC)(ip). The cultured human cancer cell line,Hela cells, was incubated with the SVC. The antineoplasiaeffects of SVC was studied in vitro and vivo. The results showed that after SVC 0. 1 mg/kg and0. 3 mg/kg had been injected for 10 days, the rate of prolongated survival time of EAC mice was52. 04% and 54. 38% respectively. The difference was significant. When the injection of SVC wasstopped, 10 days later the inhibition rate of weight was 31. 1% and 24. 2% respectively,and the dif-ference was also significant. The experiment was repeated twice. All the results showed that SVC couldprolong the survived time and inhibit the increase of the weight of EAC mice. The result of the MTTmethod indicated that SVC had obvious cytotoxic effect on EAC cells of certain concentration afterEAC cells was incubated with SVC in vitro. But SVC showed no apparent effect on Hela cells.
