IL-10单核苷酸多态性检测及其与中国西藏藏族人群肺结核易感相关性研究

目的探讨白介素10(IL-10)基因启动子-592 C/A基因多态性在中国西南藏族肺结核人群与藏族健康人群中的分布,了解该位点与藏族肺结核易感相关性以及与C反应蛋白(CRP)产生水平之间的关联。方法应用限制性片段长度多态性-聚合酶链反应(PCR-RFLP)和琼脂糖凝胶电泳技术检测169例藏族肺结核患者和150例藏族健康对照者,Th2代表性细胞因子IL-10-592C/A位点,检测其基因型、等位基因频率,检测肺结核组的CRP水平并按照CRP水平异常与否进行分组比较,分析其差异。结果 IL-10-592位点AA基因型和A等位基因在藏族肺结核组中的频率显著高于对照组,其差异有统计学意义(P﹤0.05),OR=1.326,95%CI(1.142~1.531);IL-10-592C/A位点基因型在不同血清CRP水平的组间差异有统计学意义(P﹤0.05)。结论 L-10-592C/A的A等位基因是藏族人群肺结核发生相关的危险因素,AA基因型可能与藏族肺结核易感相关,其基因多态性与血清CRP水平异常相关。

Study on Association Between Interleukin-10 Gene Polymorphism with pulmonary tuberculosis in Tibetans

OBJECTIVE To investigate the relationship between the polymorphism of interleukin-10 gene(IL-10)-592C/A with the lever of CRP and susceptivity of Pulmonary tuberculosis in Tibetans.METHODS 169 Tibetan PTB patients and 150 Tibetan controls were enrolled to this study.Their IL-10-592C/A genotyping was detected by PCR-restriction enzyme analysis.RESULTS IL-10-592 A/A genotype and A allele frequency in PTB were significantly higher than those in healthy controls(P﹤0.05,OR=1.326,95%CI(1.142-1.531).CRP levels were significantly increased among PTB with AA genotype of IL-10 gene polymorphism compared to patients with other genotypes(P﹤0.05).CONCLUSION IL-10-592 A allele was the independent risk factor for PTB occurrences.Furthermore,AA genotype was related to the risk of PTB,and is related to the CRP level with tuberuculosis patients in Tibentan..
