三亚市食物中毒中人感染副溶血性弧菌的毒力基因及耐药性研究

摘    要:目的 了解三亚市食物中毒中人感染副溶血性弧菌携带毒素基因及耐药性情况,为副溶血性弧菌食物中毒病人的治疗提供参考依据.方法 利用19种不同的抗生素药敏纸片,采用K-B扩散法进行耐药检测;用荧光定量PCR扩增法,以检测菌株的TDH和TRH基因.结果 经检测的30份菌株,28株携带有TDH基因,阳性率93%,所有菌株均没携带TRH基因.30份菌株对诺氟沙星、头孢曲松、氧氟沙星、头孢哌酮、庆大霉素、氯霉素、头孢克洛、氨苄青霉素、妥布霉素、环丙沙星、洛美沙星、依诺沙星、亚胺培南、萘啶酸未见有耐药株;青霉素G、苯唑西林、羧苄青霉素全部耐药;头孢噻吩、链霉素部分耐药.结论 带有TDH毒力基因是三亚市近年来引起食物中毒中人感染副溶血性弧菌主要分子流行病学特征,所分离菌株对大多数常见抗生素敏感性高.研究结果为评价引起食物中毒的副溶性弧菌的分子流行病学特征和制定可行的防控措施提供了科学依据.

Abstract:OBJECTIVE To understand the gene of Vibrio parahemolyticus and its drug resistance,so as to provide reference for clinical therapy of patients.METHODS Kirby-Bauer disc diffusion method was employed to perform antimicrobial susceptibility testing by using 19 kinds of paper discs.The assay for tdh and trh genes was carried out by utilizing quantitative real time PCR.RESULTS Among 30 isolated strains of bacteria,28 of them carried TDH gene,with the positive rate of 93%.No trh gene was detected from all strains of bacteria.Drug resistance was not observed in the 30 strains if bacteria,including norfloxacin,ceftriaxone,ofloxacin,cefoperazone,gentamycin,chloromycetin,cefaclor,ampicillin,tobramycin,ciprofloxacin,lomefloxacin,enoxacin,imipenem and nalidixic acid.All isolates were resistant to penicillin G,oxacillin and carbenicillin.Some of the isolated bacteria showed resistance to cephalothin and streptomycin.CONCLUSION Tdh gene of Vibrio parahemolyticus isolated from food-poisoning cases has become one of the major molecular epidemiologic characteristics in recent years in Sanya.Isolated bacteria in the present study exhibited antimicrobial susceptibility to most commonly used antibiotics.The results provided scientific ground for evaluation on the molecular epidemiologic characteristics and making feasible strategies and measures for control and prevention.
