摘    要:目的探讨CD40-CD40配体系统(CD40-CD40L)对不同因素致动脉粥样硬化形成的影响及机制研究。方法 25只日本大耳白兔普通颗粒饲料喂食1周后,随机分为5组,每组5只。A组(正常对照组):普通颗粒饲料喂饲;B组(高脂模型组):高脂饲料喂饲;C组(高脂模型CD40阻断组):高脂饲料喂饲同时给予抗CD40配体抗体0.5ml(250μg),腹腔注射,每周两次;D组(免疫刺激组):应用肺炎衣原体感染制备动脉硬化模型;E组(免疫刺激CD40阻断组):制备模型的同时给予抗CD40配体抗体0.5ml(250μg),腹腔注射,每周两次;实验共8周。实验结束后,检查如下指标:①血脂、hsCRP。②ELISA法的检测sCD40L。③观察主动脉组织病理形态学改变。8周末实验结束后获取血浆和主动脉全长标本进行生化和病理分析。结果 B、C、D、E组与A组比较,血浆TC、TG、LDL-C升高,HDL-C水平降低(P﹤0.01)。B、C、D、E组与A组比较高敏C-反应蛋白(hsCRP)水平升高(P﹤0.01)。C、E组与B、D组比较hsCRP、sCD40L浓度明显降低(P﹤0.01)。B、C、D、E组主动脉内膜有不同程度的脂质斑块形成,B组脂质斑块最多。结论通过高脂饮食、免疫刺激可引起动脉粥样硬化斑块形成及sCD40L水平升高,阻断CD40-CD40L可以抑制炎症反应,减少动脉粥样硬化斑块面积,减轻动脉粥样硬化的进展。

Abstract:OBJECTIVE To observe the mechanism of CD40-CD40L in atherosclerosis.METHODS A total of 25 Japanese big ear rabbits were randomly divided into 5 group after ordinary pellet feed for 1 week.Group A:Ordinary pellet feed;Group B:High fat diet feed;Group C:High fat diet feed with CD40 ligand antibody given twice a week(0.5ml);Group D:Arteriosclerosis model by chlamydia pneumoniae infection;Group E:Arteriosclerosis model with CD40 ligand antibody given twice a week(0.5ml).8 weeks later,we checked blood lipids,hsCRP,sCD40L,and pathological changes of aortic tissue.RESULTS Compared to Group A,the TC,TG,LDL-C level of Group B,C,D,E were raised,and the HDL-C level decreased(P﹤0.01).Compared to Group A,the hsCRP level of Group B,C,D,E were raised(P﹤0.01).Compared to Group B and D,the concentration of hsCRP,sCD40L in Group C,E were decreased(P﹤0.01).Group B,C,D,E had different levels of lipid plaque formation aortic intima.Group B had the most lipid plaque.CONCLUSION High fat diet and immune stimulation are risk factors of atherosclerosis and can raise sCD40L level.CD40 ligand antibody can inhibit inflammatory response,decrease the area of atherosclerosis and reduce progression of atherosclerosis.
