术后疲劳综合征动物模型的研究

摘    要:目的:建立并评价肝部分切除致大鼠术后疲劳综合征实验动物模型(Postoperative Fatigue,POF大鼠模型)。方法:根据POF的发病特征与临床表现,模仿手术的全过程,切除大鼠的部分肝脏,观察手术前后大鼠的行为学改变、营养学指标变化及小肠粘膜应激损伤等,建立并评价POF大鼠模型。结果:不同比例肝部分切除术后大鼠出现不同程度的术后疲劳综合征表现:眯眼、动物体力下降、活动不能、出现抑郁倾向;松毛、竖毛及毛发无光泽;肛周、眼周及鼻周污物附着并表现为体温低、肌肉寒颤及四肢冰冷等,对针刺足跖的敏感性反应降低;同时在营养学指标、小肠粘膜应激损伤等发生变化。其中以70%肝切组大鼠变化最为明显。结论:肝部分切除致大鼠术后疲劳综合征实验动物模型的建立,其最适合的切肝量为70%,该模型可作为今后POF研究的药理实验动物模型。

Abstract:Objective:To establish and assess the model of post operative fatigue syndrom(POF) by partial resection of the liver in rat.Methods:The rat model of POF was developed by the partial resection of the liver and those behavioral changes prior and post to operation,the disorder of nutritive intake after operation,stress reaction,(pathological change of mucous membrane in small intestine) would be observed.Results:Signs such as narrowing eyes,falling in physical strength and activity,tendency to depression,erecting hairs and the loss of luster in hair dirty attachment around annuls,eyes and noses low temperature,phricasmus and ice-cold extremities and lower sensitivity and so on,as the index of nutrition obviously dropped in 70% liver resected rats.The injury of the mucous nenbrane was resulted from the stress reaction after the resection of the liver.Conclusion:The similar signs to the clinical patients with POF are seen in the rat model by 70% resection of the liver,it suggests that the best proportion of the resection is 70%.The model can be used for pharmacological test.
