3种血清葡萄糖测定方法的比较

摘    要:目的比较葡萄糖氧电极法和葡萄糖氧化酶法以及己糖激酶法3种测定血清葡萄糖方法之间的灵敏度、线性、精密度以及相关性,用于指导临床实验。方法采用Beckman Coulter Synchron CX3的葡萄糖氧电极法和Modular PPI的葡萄糖氧化酶法以及己糖激酶法3种方法,同时对相同的标本进行检测比较。结果 3种方法之间相关性良好。葡萄糖己糖激酶法线性范围大于氧电极法大于氧化酶法。葡萄糖氧化酶法,己糖激酶法精密度均高于氧电极法,己糖激酶法、氧化酶法的批内变异小于葡萄糖氧电极法,而二者批间变异则大于葡萄糖氧电极法。溶血对葡萄糖己糖激酶法、氧电极法有影响,对葡萄糖氧化酶法无明显影响。结论 3种方法均符合临床对血清葡萄糖检测的要求,但葡萄糖氧电极法准确﹑简便﹑快速﹑范围较宽,应是未来血清葡萄糖测定的发展方向。

Abstract:Objective To compare the sensitivity,linearity,precision and correlation of the glucose oxygen electrode method,glucose GOD-PAP method and glucose hexokinase(HK) method of serum glucose detection for guiding the clinical tests.Methods The same specimens were simultaneously detected by the glucose oxygen electrode method on the Beckman Coulter Synchron CX3,and the glucose GOD-PAP method and glucose HK method on the Roche Modular PPI.Then the detection results were compared A.Results The correlation among three methods was better.The linear range of the glucose HK method was wider than that of the glucose oxygen electrode method and the glucose GOD-PAP method.The precision of the glucose HK method and the glucose GOD-PAP method both were higher than that of the glucose oxygen electrode method.The intra-assay CV of the glucose HK method and the glucose GOD-PAP method was less than that of glucose oxygen electrode method.But inter-assay CV of the glucose HK method and the glucose GOD-PAP method was higher than that of the glucose oxygen electrode method.Hemolysis had influence on the glucose HK method and the glucose oxygen electrode method,but no influence on the glucose GOD-PAP method.Conclusion The three methods all conform to the clinical requirement for glucose detection.The glucose oxygen electrode method is accurate,simple and rapid with wide linear range,which may be the developing direction of serum glucose measurement in the future.
