替米沙坦对博来霉素诱导的大鼠肺纤维化模型的干预作用

摘    要:目的研究替米沙坦对博来霉素诱导的大鼠肺纤维化模型的干预作用,并探讨其机制。方法雌性SD大鼠45只随机等分为三组:生理盐水对照组、博来霉素组(气管内注入博来霉素)及替米沙坦组(气管内注入博来霉素,并给予替米沙坦灌胃)。采用HE染色了解肺泡炎和肺纤维化的情况,酸解法测羟脯氨酸含量,ELISA检支气管肺泡灌洗液(BALF)中转化生长因子-β1(TGF—β1)水平的变化,并观察BALF中的细胞总数变化。结果替米沙坦组与博来霉素组比较,灌注后第7和14天肺泡炎轻;第28天肺纤维化减轻。博来霉素组肺羟脯氨酸含量增高,替米沙组减少。替米沙坦组BALF中的细胞总数第7和14天较博来霉素组少。替米沙坦组BALF中TGF—β1水平于各时间点低于博来霉素组。结论替米沙坦对博来霉素诱导的大鼠肺纤维化模型有抑制作用,其机制可能与抑制TGF-β1水平有关。

Effects of Telmisartan on pulmonary fibrosis of rats induced by bleomycin

Abstract:Objective To investigate the effects of Telmisartan on bleomycin-induced pulmonary fibrosis in rats and its possible mechanism. Methods Forty-five female SD rats were randomly divided into three groups (n = 15 in each group), bleomycin group(intratracheal injection with bleomycin), Telmisartan group(intratracheal injection with bleomycin and fed with Telmisartan via a gastric tube), and control group. The rats were sacrificed at 7, 14 and 28 days after the treatment. The lung were harvested for histopathologic HE staining. The expression of HYP in the lung was examined. The levels of transforming growth factor-β1 (TGF-β1)in bronchoalveolar lavage fluid (BALF) were assayed by enzyme-linked immunosorbent analysis. The number of total cells in BALF was also counted. Results In bleomycin group, the alveolus inflammation and pulmonary fibrosis were apparent. In Telmisartan group, however, the alveolus inflammation and pulmonary fibrosis were much milder. The expression of HYP in lungs was reduced and the the number of total cells in BALF was significantly inhibited by Telmisartan. Overexpression of TGF-β1 in BALF on day 7, 14 and 28 day induced by bleomycin was also suppressed significantly by Telmisartan. Conclusion The pulmonary fibrosis induced by bleomycin can be inhibited by Telmisartan to some extent. The inhibitory effect of Telmisartan may be ascribed to the reduction of TGF-β1 partly.
