摘    要:探讨IL-4基因修饰对K562细胞诱导的肿瘤特异性CTL杀伤活性的促进作用及可能机理。采用逆转录病毒体LXSN将IL-4 cDNA导入K562细胞,经G418筛选后获得表达IL-4蛋白的K562克隆。以野生型和载体修饰的相应细胞为对照,检测了IL-4基因修饰K562细胞诱导的T细胞增殖反应,用^51铬(^51Cr)释放法检测CTL杀伤活性。结果发现IL-4基因修饰的K562细胞诱导的T细胞增殖反应

Abstract:To observe the effect and possible mechanism of IL-4 gene modification on killing activity of K562 -specific CTL, IL-4 cDNA was transduced into K562 cell via the retroviral vector-mediated gene transfer method. G418-resistant clones were obtained and expression of IL-4 of IL-4 gene-engineered and parental K562 cells was determined. Meanwhile, proliferation of normal PBMC cells were assayed by 3H-TdR incorporation and the 51Cr release test, respectively. The results showed that proliferation of normal PBMC and killing activity of CTL induced by IL-4 positive K562 cells were greatly increased. It was concluded that IL-4 gene modification is capable of upregulating the killing activity of K562-specific CTL by possibly enhancing the T lylnphocyte function.
