摘    要: 目的:制备丝裂霉素C铁磁性微球(FMS),探讨工艺条件对成球的影响,并測定微球粒径、组分含量及体外 药物溶出度等主要物理性质。方法:以人血清白蛋白作载体,純铁粉作磁响应物质,用加热固化法制备包埋抗肿瘤 葯物丝裂霉素C(MMC)的铁磁性微球,显微镜及电镜观察微球形态、粒径分布,用原子吸收分光光度法測定其中 铁含量,紫外分光光度法測定MMC含量及体外药物溶出度。结果:乳化时间、搅拌速度、白蛋白投料比对微球粒径 影响很大,控制适当的工艺条件可得粒径为20-100μm,铁磁性物质含量为22.4%-24.3%,药物含量适中,且具 有一定緩释性能的磁性微球。结论:自制的FMS符合临床使用要求,有希望作为新型药物载体应用于靶向给葯系 统。

Abstract:OBJECTIVE To prepare ferromagnetic mitomycin C-bearing microspheres (FMS) investigate technological factors of microsphere formation,and determine physical properties such as size,contents of Fe and mitomycin C(MMC) and dissolution rate of FMS in vitro. METHODFMS were prepared by heat denaturation using human serum albumin as a matrix and pure powdered Fe as magnetically responsive material. The shape and size of FMS were observed with microscope and scanning electron microscope. Magnetic content was determined by atomic absorption spectroscopy. The percentage incorporation of MMC and the dissolution rate were assessed by UV spectrophotometry. RESULTSEmulsifying periods,stirring speeds and albumin percent have great influence on size and distribution of FMS. Under proper conditions,FMS in diameter of 20-100 pm,containing 22.4%?24.3% Fe and 0.31% MMC was prepared. The drug entrapped in FMS could be released sustainedly. CONCLUSION The microspheres prepared in our laboratory could be expected to apply to clinical targeting system as a carrier of antineoplastic drug.
