RNA干扰中小干扰RNA的设计原则

摘    要:RNA干扰(RNAi)是由内源性或外源性双链RNA介导的一种高度保守的序列特异性基因表达调控机制,主要通过小干扰RNA(siRNA)和微小RNA发挥作用。由于其具有高效、高特异性、作用稳定等特点,已被广泛地应用于基础实验和疾病治疗的研究中。在进行siRNA介导的RNAi实验时,如何设计高效、特异性抑制靶mRNA的siRNA是实验成败的关键。

Abstract:RNAi interference(RNAi)is a highly conserved and sequence-specific regulatory mechanism of gene expression induced by exogenous or endogenous,double-stranded RNA,primarily by small interfering RNA(siRNA)and microRNA(miRNA).Due to its high efficiency,high specificity and good stability,RNAi has been widely used in the research of basic experiment and treatment of disease.During the RNAi experiment induced by siRNA,how to design the efficient and specific siRNA is the key of a successful experiment.
