异甘草素和Flutamide联合应用对人前列腺癌细胞体外增殖的抑制作用

摘    要:目的观察异甘草素(ISL)和Flutamide联合应用对人前列腺癌细胞体外增殖的抑制作用。方法采用MTT法测定细胞增殖,流式细胞仪检测细胞凋亡,逆转录聚合酶链反应(RT-PCR)分析Bcl-2表达。结果ISL和不同浓度Flutamide联合用药比单独应用Flutamide对前列腺癌PC-3细胞增殖抑制作用明显增强(P<0.01),同时显著下调Bcl-2基因表达,增加癌细胞凋亡。结论ISL和Flutamide联合应用对前列腺癌PC-3细胞增殖抑制作用明显增强,其机制可能与下调Bcl-2基因表达及增加癌细胞凋亡有关。

Abstract:Objective To study the effect of isoliquiritigenin(ISL)liposome combined with Flutamide on the prolifera- tion of human prostate cancer cells in vitro.Methods The cell proliferation was evaluated by MTT method.Cell cycle and apoptosis were determined by flow cytometer(FCM) and RT-PCR was used to detect the expression of Bcl-2.Results ISL combined with Flutamide showed a significantly stronger inhibitory effect on PC-3 cell in vitro than Flutamide alone(P<0.01). The expression of Bcl-2 was down-regulated and the apoptosis of PC-3 cell was increased also.Conclusion Proliferation inhibitory effect against PC-3 cell in vitro can be enhanced by ISL combined with Flutamide.The possible mechanism is associated with increasing apoptosis of PC-3 cell and decreasing expression of Bcl-2.
