CD8类McAb—SM4的研制与鉴定

本文报导一株抗人T细胞亚群单克隆抗体SM_4的研制与鉴定。SM_4仅与T细胞中的一个亚群反应,在淋巴组织冰冻切片上,SM_4只与胸腺依赖区的部分细胞反应,其组织分布特征与CD_8单抗相类似。叠加试验结果表明SM_4与CD_8类单抗识别同一细胞亚群——Ts/Tc亚群。竞争抑制试验进一步证明SM_4识别CD_8分子上某一特定表位。本文还就CD_8亚群异质性问题及SM_4的应用潜力进行了讨论。

Production and Identification of Anti-Human T Lymphocyte McAb (SM4) Recognizing A Epitope of CD8 Molecule

A monoclonal antibody (McAb) against a subset of human T lymphocytes was produced and identified. The McAb,named SM4,reacted exclusively with cells of T lineage including thymocyte,peripheral blood T cells and T cell lines. It gave negative reaction with peripheral blood B cells,granulocytes,red blood cells,B cell lines and other cell lines. The histochemical distribution of SM4 on cryostat sections of lymphoid tissues was similar to that of CD8-like McAbs. Then comparison of SM4 with standard CDS-like McAbs has been done. The result of additive test demonstrated they reacted with the same T subset-Ts/Tc subset,and the competitive blocking test showed SM4 recognized a particular epitope of CD8 molecule. The heterogeneity of CD8 subset and the potential applificatiou of SM4 were discussed.
