银杏叶提取物防治兔心肌缺血再灌注损伤实验研究

摘    要:目的 探讨家兔心肌缺血再灌注损伤中血小板活化因子 (Platelet-activatingfactor,PAF)与血小板活化因子 -乙酰水解酶(PAF -Acetyl-hydrolase,PAF -AH)的变化以及银杏叶提取物 (ExtraGinkgoBiloba ,EGB)对家兔心肌缺血再灌注损伤的防治效果。方法 取 30只家兔随机分为 3组 :正常家兔组 (n =1 0 ) ;缺血再灌注对照组 (n =1 0 ) ;EGB治疗缺血再灌注组 (n =1 0 ) ,按文献建立模型 ,检测各组PAF、PAF -AH、丙二醛 (MDA)和ATP的变化。结果 缺血再灌注对照组与EGB治疗缺血再灌注组PAF含量显著高于正常家兔组 (P <0 .0 1 ) ,而PAF -AH的活性显著低于正常家兔组 (P <0 .0 1 ) ;缺血再灌注对照组心肌组织MDA含量显著高于EGB预处理组 (P <0 .0 1 ) ,而ATP的含量显著低于EGB预处理组 (P <0 .0 1 )。结论 PAF与AF -AH的变化在心肌缺血再灌注损伤中发挥重要作用 ,EGB对心肌缺血再灌注损伤有一定的防治作用 ,其机制与EGB阻断PAF -R等因素相关。

Abstract:Objective To study the roles of plasma PAF and PAF-AH and the preventive and therapeutic effects of Extra Ginkgo Biolba (EGB) on the injury caused by myocardial ischemia reperfusion in rabbits. Methods 30 male rabbits were randomly divided into 3 groups: normal group (n=10); the model myocardial ischemia reperfusion control group (n=10); EGB treatment group (n=10). Models were set up according to literature, and the changes of PAF,PAF-AH, MDA and ATP in the respective groups were tested. Results The concentration of PAF in the control group and EGB group was higher ( and the activation of PAF-AH was lower) than that in the normal group with significant statistical difference (P<0.01). The concentration of cardiac muscle MDP in the control group was higher ( and the concentration of cardiac muscle ATP was lower) than that in EGB group again with significant statistical difference (P<0.01). Conclusion The data indicate that the changes of PAF and PAF-AH are related to the occurrence and development in the injury of myocardial ischemia reperfusion; EGB has preventive and therapeutic effects in injury of myocardial ischemia reperfusion.
