淋球菌膜蛋白表达与环丙沙星耐药性的相关性研究

摘    要:目的:探讨淋球菌细胞膜蛋白表达与环丙沙星耐药性的相关关系。方法:采用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)和直接荧光法分别检测淋球菌外膜孔蛋白和内膜泵蛋白的表达以及淋球菌对环丙沙星的摄入蓄积量。结果:耐药菌对环丙沙星的蓄积量明显低于敏感菌;敏感菌外膜上均有33kd孔蛋白的表达,而耐药菌外膜上则缺失33kd蛋白;但大部分耐药菌内膜上却存在泵蛋白的表达。结论:淋球菌外膜孔蛋白表达的缺陷及内膜泵蛋白的表达使淋球菌对环丙沙星的蓄积量明显降低,从而导致淋球菌耐药。

Abstract:Objective To explore the correlation between protein expression in cell membrane of neisseria gonococci and resistance of gonococci to ciprofloxacin(CIP).Methods We used the methods of SDS-PAGE respectively to detect the expression of porin protein in the outer membrane and efflux pump in the inner membrane of these gonococci.Also we determined the uptake and accumulation of ciprofloxacin in neisseria gonorrhoeae by direct fluorescence assay.Results The accumulation of CIP in resistant strains were obviously lower than that in sensitive strains.The 33kd porin was expressed in the outer membrane of sensitive strains,but not in the outer membrane of resistant strains of gonococci,there were protein expression of efflux pump in the inner membrane of most resistant strains.Conclusion The defect of porin protein expression in the outer membrane of gonococci and the efflux pump expression in the inner membrane obviously decrease the accumulation of CIP in gonococci and induce the resistance to CIP.
