培哚普利对盐敏感性高血压大鼠心脏形态及其功能的影响

摘    要:目的 观察盐敏感性高血压动物模型之心脏形态及功能的变化 ;培哚普利 (雅施达 )对该模型心脏重塑的干预作用 :并对它们的作用机理作初步探讨。方法 新生雄性 Wistar大鼠在第 1d和第 2 d分别皮下注射辣椒辣素以建立感觉神经损伤性盐敏感性高血压大鼠模型 ,以培哚普利治疗 4个月前后及与同龄对照鼠比较 ,进行有创心功能检测、心肌病理形态学检查。结果  1.注射辣椒辣素和高盐饮食可形成盐敏感性高血压动物模型。 2 .该模型心脏重塑表现为 :心肌细胞短径增大、排列紊乱、间隙缩小 ,心肌间质胶原成分增多 ;左心室收缩峰压 (L VSP)、左心室舒张末压 (L VEDP)、等容舒张期室内压下降的时间常数 (T)均增高。 3.经培哚普利治疗后 ,该模型鼠尾收缩压降低 ,心功能、心肌病理形态学指标均有改善。结论  1.辣椒辣素处理的新生大鼠在高盐饮食时可迅速建立起一种盐敏感性高血压模型。 2 .该模型存在心肌重塑现象 :心肌细胞肥大、心肌间质胶原增多、心肌舒张功能下降。 3.培哚普利能有效阻止盐敏感性高血压及其心脏重塑的产生 ,并保护心功能。

Abstract:Objective To observe changes of cardiac morphology and its function; To observe the interferential effects of cardiac remodeling after using perindopril (acertil)in the salt-sensitive hypertensive rat model.MethodsTo establish salt-sensitive hypertensive rat model induced by sensory denervation, newborn male Wistar rats were given capsaicin subcutaneously on the 1st and 2nd day of life. Comparisons of before and after fourmonths therapy of perindopril (acertil) and of salt-sensitive hypertensive rats and match-age rats were observed.After four months,invasive cardiac function and cardiac muscle morphology were examined. Results 1. Development of salt-sensitive hypertensive rat model induced by sensory denervation was successfully copied by injected capsaicin and high salt diet.2. cardiac remodeling of the model represented as follows:the parameters of myocyte short diameter, cardiac muscle fibrillar collagen,left ventricular systolic pressure(LVSP),left ventricular end-period diastolic pressure (LVEDP) and time constant (T) were significantly higher than controls'. Disorganized array of cardiac cells and decreased interspace among cardiac cells were observed. 3.After treatment,the above referencial indices were all improved. Conclusion 1. The salt-sensitive hypertensive rat model was established successfully when neonatal Wistar rats treated with capsaicin was fed a high salt diet. 2.Cardiac remodeling and abnormally cardiac function were observed in the model.3. Acertil could effectively prevent blood pressure from raising and keep left ventricle from remodeling and be beneficial to cardiac funtion.
