摘    要:新近研究表明丝裂素活化蛋白激酶(MAPK)是细胞增殖肥大的重要酶类,为各种细胞外的生长刺激信号引起细胞内信息传递的共同通路或汇聚点。本文通过观察自发性高血压大鼠(SHR)心肌MAPK活性和心肌肥大的关系,探讨SHR心肌肥大发生的可能细胞内信息传递机制。方法:4个月的SHR和WKY大鼠各8只,以心重/体重的比值表示心肌肥厚的程度,采用胶内磷酸化法测定心肌MAPK活性。结果:与WKY大鼠比较,SHR心肌MAPK活性增加107.0%(P<0.01),心肌肥大程度增加38.3%(P<0.01)。心肌MAPK活性与心肌肥大程度呈显著正相关(r=0.708,P<0.05)。结论:SHR心肌肥厚可能涉及MAPK。

Abstract:Mitogen-activated protein kinases (MAPKs) are thought to be critical components in signal transduction pathways which regUlate cell growth and differentiation. It has been suggested that the MAPK may be represented a convergence point Where diverse signaling pathways are brought together. In an attempt to assess a possi-ble involement of MAPK in the development of cardiac hypertrophy in spontaneously hypertensive rats (SHR), we observed the relationship between Cardiac hypertrophy and MAPK activity in Cardiac tissue. Method: The animals were divided into two groups: 4 months old SHR (n = 8) and WKY rats (n = 8). The ratio of heart to they weight was measured. MAPK activity in Cardiac tissue was assayed by the "in - gel" myelin basic protein phosphorylation. Results: Compared with WKY rats, MAPK activity in cardiac tissue of SHR increased 107. 0% (P< 0. 01) with a marked cardiac hypertrophy (P < 0. 01). There was a positive correlation between MAPK activity and Cardiac hypertrophy (R=0.708, P<0.05).
