高侵袭力人膀胱癌细胞亚系的分离鉴定

摘    要:目的建立BIU-87、EJ和T24 3株膀胱癌细胞的高侵袭力亚系,并对侵袭过程进行观察。方法通过构建侵袭小室,以穿透人工基底膜、聚碳酸脂膜能力为依据筛选具有体外高侵袭能力的膀胱癌细胞亚系。运用扫描电镜观察细胞侵袭的过程,通过生长曲线、细胞周期、体外侵袭力对母代与子代细胞体外增殖及侵袭能力进行对比。通过裸鼠膀胱灌注不同侵袭力的肿瘤细胞,对侵袭力与种植转移的关系进行初步研究。结果通过扫描电镜观察到了细胞的侵袭运动。筛选后的各子代细胞,经16代以上传代后,细胞形态与母代无明显变化,细胞增殖速度较母代有明显加快(P<0.05),体外侵袭能力较母系明显提高(P<0.001)。结论运用改良的侵袭实验筛选膀胱癌细胞系的高侵袭力亚系的方法是可靠的,建立的高侵袭力亚系细胞为今后对膀胱癌侵袭及转移的研究提供了较为可靠的细胞模型。

Abstract:Objective To establish the high invasiveness subseries cell lines of three bladder neoplasm strains of BIU- 87, EJ and T24, and to observe the invasive process in vitro. Methods The high invasiveness subseries cell lines were selected based on the penetrating power of artifieal basilar membrane and polyearbonate membrane through constructing invasive cabin. The invasive process were observed by scanning electron microscope. The penetrating power and cell proliferation power in vitro of mother generation and filial generation were compared by growth curve, cell cycle and invasiveness in vitro. The relationship between invasiveness and implantation metastasis was studied by irrigation of bladder tumor cells with different invasiveness. Results Invasive movement of the cell had been observed through scanning electron microscope. After the sixteenth transfer of culture, compared with mother generation, there was no marked change in cell morphous of filial generation, the velocity of cell proliferation obviously speeded up( P 〈 0.05 ), invasiveness in vitro obviously increased ( P 〈 0.001 ) and the rate of forming tumor of filial generation with high invasiveness was higher than that of mother generation. Conclusion It is reliable method to construct subseries cell lines of bladder neoplasm strains with improved invasive experiment, and the subseries cell lines can be used as cell model for studies of bladder eareinoma's invasiveness and metastasis.
