摘    要:目的:应用Box-Behnken试验设计优选黄芩的超高压提取工艺。方法:以黄芩苷提取率为指标,采用单因素试验考察提取压力、提取次数、液固比等因素对黄芩超高压提取工艺的影响;Box-Behnken试验设计考察乙醇体积分数、液固比、提取压力对提取工艺的影响,优化工艺参数。结果:最佳提取工艺条件为乙醇体积分数72%,提取压力100 MPa,液固比100∶1,提取时间10 s,黄芩苷提取率102.65 mg.g-1。结论:优选的工艺提取率高、提取温度低、耗时短、能耗少,适用于黄芩的工业化提取。

Abstract:Objective:To optimize ultrahigh pressure extraction (UPE) process of baicalin from Scutellaria baicalensis by Box-Behnken design. Method: With yield of baicalin as index,influence of extraction pressure,extraction times,liquid-solid ratio and other factors on UPE of S. baicalensis was investigated by single factor test;Box-Behnken design was employed to optimize extraction technology parameters with ethanol concentration, liquid-solid ratio and extraction pressure as independent variables. Result: Optimum extraction technology conditions were as following:extracted 10 s with 100 times the amount of 72% ethanol by extraction pressure of 100 MPa,yield of baicalin was 102.65 mg·g-1. Conclusion: Optimized technology had higher yield,lower extracting temperature, shorter extacting time and less power consumption,it was suitable for industrial production of S. baicalensis.
