黄荆子乙酸乙酯提取物体内抗炎作用及机制

摘    要:目的研究黄荆子乙酸乙酯提取物(EVn-50)体内抗炎的作用效果及其可能的机制。方法建立四种急性炎症模型,二甲苯致小鼠耳廓肿胀模型,鸡蛋清致大鼠足跖肿胀模型,腹腔注射0.7%醋酸致小鼠腹腔毛细血管通透性增高模型和腹腔注射去甲斑蝥素悬液致大鼠全身性炎症模型。观察不同浓度的EVn-50对急性炎症模型的抑制作用,用western blot检测全身性炎症大鼠模型血中白细胞p38MAPK蛋白表达情况,用ELLSA检测全身性炎症大鼠模型血清TNF-α、IL-1、IL-6、COX-2炎性细胞因子表达情况,探求EVn-50抗炎可能机制。结果EVn-50对四种急性炎症模型均有显著抑制作用,与NS组比较,差异有统计学意义(P〈0.05),同时去甲斑蝥素诱导的全身性炎症大鼠血液白细胞p38MAPK蛋白表达降低,TNF-α、IL-1、IL-6、COX-2炎性细胞因子表达减少。结论 EVn-50可以显著抑制各种急性炎症反应,其机制可能是通过抑制p38MAPK细胞信号转导通路实现的。

Effect of EVn-50 on anti-inflammation in vivo and its mechanism

Abstract:Objective The aim of this study is to investigate the anti-inflammatory effects of ethylacetate extracts of vitex negundo L.(EVn-50) in vivo and its possible mechanism.Methods The acute inflammation models were established including xylene-induced mouse ear edema,egg albumen-induced rat paw edema,acetic-induced mouse vascular permeability and norcantharidin-induced rat systemic inflammation.The anti-inflammatory effect on different concentrations of EVn-50 were observed,and the p38 MAPK protein of white blood cell were detected using western blot.The cell factors of TNF-α,IL-1,IL-6 and COX-2 of rat serum from norcantharidin-induced systemic inflammation model rat were also detected using ELISA method.Results There were significantly difference between different concentrations of EVn-50 groups and NS group in four types of acute inflammation model(P〈0.05).Meanwhile,the p38 MAPK protein of white blood cell and the cell factors of TNF-α,IL-1,IL-6,COX-2 of the serum of norcantharidin-induced systemic inflammation were down-regulation.Conclusion EVn-50 could significantly inhibit acute inflations models,and its mechanism may be related to suppress p38 MAPK signal transduction pathway.
