| 摘 要： | 目的探讨生长抑素类似物奥曲肽对人肝癌细胞株MHCC97-L中血管内皮生长因子(VEGF)mRNA表达及VEGF合成与分泌的影响.方法利用酶联免疫吸附法、流式细胞仪、逆转录-聚合酶链反应检测不同剂量的奥曲肽对人肝癌细胞株MHCC97-L的VEGF合成与分泌及VEGF mRNA表达的影响.结果奥曲肽(10-5～10-10 mol/L)可以抑制人肝癌细胞株 MHCC97-L中VEGF mRNA表达及VEGF合成与分泌,呈剂量依赖性,可见饱和现象.结论奥曲肽能够抑制人肝癌细胞株MHCC97-L中VEGF的表达、合成和分泌,可能是奥曲肽抗血管新生的作用机制之一.

| Abstract: | Objective To investigate the effects of somatostatin analogue octreotide on vascular endothelial growth factor (VEGF) expression,production,and secretion by hepatocellular carcinoma (HCC) cell line MHCC97-L. Methods The effects of octreotide on VEGF mRNA expression,and intracellular protein production and secretion in HCC cell line MHCC97-L were measured by RT-PCR,flow cytometry and enzyme linked immunosorbant assay (ELISA). Results Octreotide in concentrations from 10 -5 mol/L to 10 -10 mol/L inhibited VEGF mRNA expression,intracellular production and secretion of VEGF in HCC cell line MHCC97-L in a dose-dependent manner. Conclusion Octreotide is a potent inhibitor of VEGF expression and production by HCC cell line MHCC97-L,suggesting that the inhibitory influence of octreotide on VEGF expression and production may be one of the mechanisms underlying its anti-angiogenesis effect on HCC. |
