脂肪干细胞源性神经干细胞克隆球的电镜观察

摘    要:目的观察脂肪干细胞(ADSCs)源性神经干细胞克隆球的超微结构形态。方法原代培养大鼠ADSCs,向神经干细胞诱导分化,分化为克隆球后,免疫荧光鉴定其Nestin表达,透射电子显微镜观察克隆球超微结构形态。结果大鼠ADSCs可诱导分化为细胞克隆球,其Nestin表达阳性,超微结构与原代培养的神经干细胞克隆球的超微结构相似。结论 ADSCs经诱导分化后形成的克隆球具有神经干细胞的特征。

Ultrastructure of neural stem cell neurospheres differentiated from adipose-derived stem cells under transmission electron microscope

Abstract:Objective To observe the ultrastructure of neural stem cell neurospheres differentiated from adipose-derived stem cells(ADSCs) by transmission electron microscope.Methods ADSCs were isolated from the inguinal fat of the SD rats and then ADSCs were induced to differentiate into neurospheres.immunofluorescence technic was used to detect the expression of Nestin and transmission electron microscope was used to observe the ultrastructure of neurospheres differentiated from ADSCs.Results ADSCs were successfully induced to differentiate into neurospheres.The neurospheres differentiated from ADSCs expressed Nestin and showed a similar ultrastructure to the neurospheres differentiated from neural stem cells.Conclusions neurospheres differentiated from ADSCs show some characteristic of neural stem cells.
