雌二醇对去卵巢大鼠血管内皮细胞功能的影响

摘    要:目的本实验用去卵巢大鼠建立更年期动物模型,给予不同剂量外源性雌二醇(E2),检测血清中一氧化氮(NO)和内皮素(ET)含量的变化,观察E2对心肌和血管细胞凋亡以及对心肌和血管结构形态的改变,进一步探讨E2对于心脏及血管内皮细胞功能的影响。方法用放射免疫分析方法(RIA)测定血清中E2和ET;用酶法测定血清中NO的含量;用荧光标记法观察心肌和血管细胞凋亡;用电子显微镜(EMS)观察心肌和血管超微结构。结果NO浓度随E2水平增加而升高,而ET浓度则随E2水平升高而降低,心肌和血管细胞凋亡以及超微结构损伤与B含量呈负相关。结论适量外源性E2对心血管系统的保护机制可能促进NO生成、抑制ET合成,同时抑制心肌和血管细胞凋亡及超微结构的损伤。

Abstract:Objective The present study establishes menopausal rat model with ovariectomized rats by injecting different dose of estradiol(E2) in checking the variation of the NO, ET and serum E2, and observing apoptosis and morphological structures of cardiac tissue and blood vessel, and aimed to discuss the mechanism that supply exogenous E2 protected the heart and vessel.Methods The present study analyzed the serum E2 and ET by radiated immunity analyzing (RIA), measured the contents of the NO in serum by enzymic methods, observed the apoptosis of cardiac and vessel cell by fluorescent marking methods and morphological ultramicrostructure changes by electron microscope(EMS). Results The results showed that NO concentration increased in serum with the raising of E2 concentration, ET concentration decreased with E2 increased. The more E2 concentration in serum, detected the more little visible phenomena of apoptosis and damage of cardiac tissue and blood vessel wall.Conclusions Supplying proper exogenous E2 can promote the NO and decrease the ET, meanwhile it can inhibit apoptosis of myocardial cells and impair of cardiac tissue, this indicated that the protection of exogenous E2 for cardiovascular system maybe reached through above ways.
