胰苏灵对2型糖尿病模型大鼠胰岛素抵抗作用的实验研究

目的观察胰苏灵对2型糖尿病胰岛素抵抗(IR)的改善作用,并深入探讨其机理。方法按照文献方法复制2型糖尿病 IR 模型大鼠,选血糖>11.1mmol/L,且具有超重、高血脂及高胰岛素血症的大鼠确定为实验性2型糖尿病 IR 大鼠,共46只进入实验。将46只2型糖尿病 IR 大鼠随机分为4组:模型组、胰苏灵小剂量组、胰苏灵大剂量组和二甲双胍组,治疗6周后禁食12h,分别测定空腹血糖(FBG)、血清胰岛素(INS)、血清总胆固醇(TC)、甘油三脂(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、血清游离脂肪酸(FFA)及血清和脂肪中肿瘤坏死因子(TNF-α)的含量。并计算胰岛素敏感指数(ISI)。结果胰苏灵煎剂能明显降低模型大鼠的 FBG、TG、LDL-C 及 FFA、TNF-α(P<0.05,P<0.01),提高 ISI、HDL-C(P<0.01)。结论胰苏灵能明显改善2型糖尿病的糖和脂代谢紊乱,降低 IR 因子,增加胰岛素敏感性。其作用可能通过对抗脂毒性、降低 FFA、TNF-α含量而改善胰岛素抵抗。
