黄芪诱导骨髓间质干细胞分化为神经元样细胞

目的体外定向诱导大鼠骨髓间质干细胞(MSC)分化为神经元样细胞.方法通过贴壁法分离大鼠MSC,体外扩增培养.中药黄芪定向诱导MSC分化为类神经元样细胞.光镜下观察细胞形态,免疫细胞化学检测神经细胞特异性抗原标志.结果大鼠骨髓间质干细胞可通过贴壁法成功分离并可在体外大量扩增.黄芪诱导90 min后大部分MSC转变为神经元样细胞,出现胞体和突起,免疫细胞化学染色NSE、Nestin呈阳性、GFAP阴性.结论大鼠骨髓间质干细胞可在体外诱导分化为神经元样细胞.

Mesenchymal stem cells induced with Astragale differentiate into neuron-like cells

Objective: To investigate the differentiation from rat marrow mesenchymal stem cells into neuron-like cells with Astragale. Methods: Mesenchymal stem cells were obtained from thighbone marrow of rats and expended in culture medium. The effect of the capacity of differentiation mesenchymal stem cell with Astragale which acted as neuron-like cells inducer was performed. Neuron-specific enolase (NSE), Nestin, gliall fibrilalry acaidic protein (GFAP) were detected by immunocytochemistry. Results: Rat mesenchymal stem cells were expended as undifferentiated in culture for more than 20 passages. Simple differentiation method with Astragale which can generate neuron-like cells was achieved with 90 minutes. The expression of NSE and Nestin in neuron-like cells were positive but GFAP was negative. Conclusions: Mesenchymal stem cells can be induced to differentiate into neuron-like cells with Astragale.
