自噬相关基因Beclin1和MAPLC3在急性白血病骨髓单个核细胞中的表达及其意义

摘    要:本研究旨在检测人急性白血病(AL)骨髓单个核细胞中自噬相关基因Beclin1和微管相关蛋白质轻链3(MAPLC3)的表达,并探讨其意义。采用透射电子显微镜(TEM)和RT-PCR方法检测初治、复发难治、完全缓解(CR)的急性白血病患者和正常人骨髓单个核细胞(BMMNC)中的自噬活性以及Beclin1、MAPLC3mRNA的表达。结果表明:急性白血病初治组骨髓单个核细胞自噬活性、Beclin1和MAPLC3mRNA表达分别为80%、0.68±0.18、0.24±0.06;在难治复发组分别为100%、0.79±0.09、0.30±0.07;在完全缓解组分别为40%、0.52±0.15、0.16±0.04;在正常对照组分别为20%、0.57±0.13、0.16±0.05。初治组和难治复发组自噬活性和Beclin1、MAPLC3mRNA的相对表达量均高于正常对照组,差异有统计学意义(p<0.05)。完全缓解组和正常对照组之间比较差异无统计学意义(p>0.05)。结论 :初治组急性白血病骨髓单个核细胞中自噬活性和自噬相关基因Beclin1、MAPLC3mRNA的表达均上调,在难治复发组自噬活性和自噬相关基因Beclin1、MAPLC3mRNA的表达上调尤为明显,这可能与急性白血病的发生、发展及其耐药相关。
