HPLC法测定姜黄提取物中有效成分含量

目的：建立高效液相色谱法同时测定姜黄提取物中姜黄素、一脱甲氧基姜黄素、二脱甲氧基姜黄素的含量.方法：采用C18色谱柱（150 mm×6 mm,5μm）,流动相为乙腈-水-冰乙酸（39∶57∶4）,流速为1.4 ml· min-1;检测波长：428 nm;柱温：35℃;进样量：20μl.结果：同时测定了姜黄素、一脱甲氧基姜黄素、二脱甲氧基姜黄素的含量,三者的线性范围分别为81.31～650.50 μg ·ml-1,26.93 ～215.42μg· ml-1,16.32～130.54 μg ·ml-1.平均回收率分别为100.9％,100.9％,100.6％,RSD分别为1.7％,1.2％,1.5％（n=9）.结论：该方法简便、准确、重复性好,可用于姜黄提取物中有效成分的测定.

Determination of Active Ingredients in Extracts of Curcuma Longa L.by HPLC

Objective：To establish an HPLC method for the simultaneous determination of curcumin,desmethoxycurcumin and besmethoxycurcumin.Methods：A C18 column （150 mm × 6 mm,5 μm） was used at 35℃ using the mobile phase of acetonitrile-water-acetic acid glacial （39∶ 57∶ 4） with a flow rate of 1.4 ml · min-1.The UV detection wavelength was 428mn.The injection volume was 20 μl.Results：The content of curcumin,desmethoxycurcumin and besmethoxycurcumin was simultaneously detected.The calibration curves showed good linearity within the range of 81.31-650.50 μg · ml-1 （curcumin）,26.93-215.42 μg · ml-1 （desmethoxycurcumin） and 16.32-130.54 μg · ml-1 （besmethoxycurcumin）,and the average recovery was 100.9%,100.9% and 100.6% with RSD of 1.7%,1.2% and 1.5 %,respectively （n =9）.Conclusion：The method is convenient,accurate and reproducible,and suitable for the determination of active ingredients in extracts of Curuma longa L.
