丙型肝炎高变区1合成肽体外刺激HCV感染PBMC细胞因子分泌的研究

摘    要:目的检测丙型肝炎病毒(HCV)高变区1(HVR1)模拟表位刺激自然感染患者外周血淋巴细胞后的细胞因子水平变化。方法细胞因子检测运用ELISA、流式细胞仪(FACS)淋巴细胞分类以及计算机模拟多肽抗原性。结果(1)1号和5号合成肽刺激HCV患者外周血单核细胞(PBMC)后,患者PBMC出现了明显的增殖;(2)培养上清中干扰素γ(IFNγ)、白介素4(IL-4)、白介素10(IL-10)都有不同程度的提高,白介素2(IL-2)和肿瘤坏死因子α(TNF-α)的水平并没有提高;(3)增殖细胞主要为CD4阳性淋巴细胞。结论我们设计的HCV高变区1合成肽引发的免疫因子的释放倾向于TH2类因子。

Abstract:OBJECTIVE: Test the cytokines secretion by peripheral blood mononuclear cells (PBMC) from hepatitis C patients after stimulation with highly variable region (HVR1) synthetic peptides. METHODS: ELISA was used to test the cytokines secretion, flow cytometry to group the proliferated cells, and the antigencity of synthetic peptide was predicted by the computer modeling. RESULTS: (1)There were PBMC proliferation when stimulated by HVR1 antigen synthetic peptides. (2) There was a rise of IFN-gamma, IL-4 and IL-10. But no rise of IL-2 and TNF-gamma was found. (3) The proliferated cells were mainly CD4+ lymphocytes. CONCLUSION: PMBC from hepatitis C patients tend to secret Th2 cytokines after stimulation with HVR1 designed by the authors.
