摘    要:目的研究转化生长因子β激活激酶(TAK1,MEKK7)基因在类风湿性关节炎滑膜组织的表达与活性变化,探讨其与类风湿性关节炎的关系。方法本实验采用病例对照研究分别利用RT-PCR和Western-blot方法检测类风湿性关节炎(RA)、骨性关节炎(OA)及非关节炎病人膝关节滑膜中TAK1基因mRNA和蛋白质的表达,并利用磷酸化抗体比较其活性差异,以验证TAK1与类风湿性关节炎发病的相关性。结果 TAK1 mRNA和蛋白在RA病人膝关节滑膜的表达水平明显高于OA和行膝关节镜探查术未见异常的病人,其TAK1酶活性亦高于OA和行膝关节镜探查术未见异常的病人。结论 TAK1基因与类风湿性关节炎发病发生、发展有一定的相关性。

Abstract:Objective To study the expression and activation of transforming growth factor-β activation of protein kinase-1(TAK1, MEKK7) in the knee joint synovial tissue of patients with rheumatoid arthritis(RA). Methods The case-control study, quantitative RT-PCR and Western-blot analysis were used to examine the expression and activation of TAK1 gene in patients with RA , patients with osseous arthritis(OA) and patients with no arthrits. Results The total levels of TAK1 mRNA and protein expression and the level of p-TAK1 protein in the knee joint synovial tissue were significantly higher in patients with RA than in patients with OA and with no arthritis. Conclusion The overexpression of TAK1 gene might be related to the pathogenesis of RA, which might provide a biological marker and a useful prognostic indicator for RA.
