摘    要:目的:了解放射损伤条件下骨髓细胞的生物力学特性及与细胞间相互作用的关系。方法:以6Gy^60Co照射昆明系小鼠制备动物模型,以细胞电泳法检测骨髓细胞的细胞电泳率,荧光探针法测定细胞膜微粘度,旋转法测细胞的粘附性,核孔滤膜法检测细胞的变形指数。结果:放射损伤后骨髓细胞的变形能力降低,细胞膜微粘度增加,粘附率降低,细胞电泳率降低。结论:放射线对骨髓细胞(包括基质细胞和造血细胞)的力学特性有显著影响,是放射损伤后造血功能减退的原因之一。

Abstract:Objective To explore the biodynamic properties of bone marrow cells and the relation with cell cell interaction in radiation injury mice. Methods Male Kunming mice were exposed to total body irradiation of 6Gy of gamma rays from 60 Co to establish animal model. Cell membrane electric charge, membrane fluidity, cell deformability, and cell adhesion were determined with electrophoresis, DPH probe, microporous membrane filter, and adhesion rate methods, respectively. Results The deformability, cell adhesion rate and membrane electric charge dramatically decreased, but bone marrow cell membrane microviscosity increased obviously after radiation injury at 1d, 3d and 7d. Conclusion Radiation has significant effect on the mechanic properties of bone marrow cells(including hematopoitic cells and stroma cells) after radiation injury. It may be one of reasons resulting in hematopoietic failure after radiation injury.
