摘    要:为研究三七总皂甙(PNS)及单体Rb1预处理对大鼠心肌的保护效应及其与热休克蛋白(HSP)的关系,采用在体缺血再灌注模型,实验分空白组、对照组、PNS组、Rb1组4组,心肌缺血40min再灌注30min检查心肌的超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量,用免疫组化法测缺血40min再灌120min的HSP70的表达及蛋白激酶C(ε-PKC)的含量。结果显示,与对照组相比,PNS组和Rb1组减少MDA的产生(P<0.01);同时,HSP70的表达,PNS组明显高于Rb1组及对照组(P<0.01),PNS组的ε-PKC的含量高于对照组(P<0.01)。结果表明三七总皂甙长时间处理可促进HSP70的表达,对心肌再灌注损伤有良好的保护效应。

Abstract:The purpose is to study the effect of panaxtnotoginsengsaponins(PNS) pretreatment or Rb1 on myocardial ischemia/reperfusion injury in rats and the relation among protective effect of PNS or Rb1,heat shock protein 70 and ε-PKC.Forty-two SD rats were dividrd into four group:Normal group(n=6),Control P group(n=12), PNS group(n=12) and Rb1 group(n=12).Malondialdehyde(MDA) and superoxide dismutase (SOD)contents were determined after 40 min ischemia followed 30 min reperfusion 。HSP70 and ε-PKC were detected by immunohistochemical technique after reperfusion of 120 min.The results showed that: As compared with the control,PNS snd Rb1 groups can significantly reduce MDA production and protect SOD activity(P<0.01).MDA production in PNS group was less than that in Rb1 group(P<0.01) and SOD activity in PNS group was less than that in Rb1 group(P<0.05).The HSP70 levels in PNS were higher than that of the control and Rb1 grougs (P<0.01).Compared with the normal,ε-PKC in PNS group was reduced slightly (P>0.05).In the control and Rb1 groups ε-PKC were reduced significantly (P<0.01).The results indicated that PNS can improve the expression of heat shock protein 70 in pretreated rats and has better protective effects than that of Rb1 against myocardial ischemia/reperfusion injury.
