宁夏栽培柴胡高效液相色谱指纹图谱的研究

摘    要:目的建立宁夏栽培柴胡高效液相色谱指纹图谱,为柴胡的鉴定及质量控制提供实验依据。方法采用HPLC—UV方法对宁夏不同产区、不同采收期的l0批柴胡药材建立指纹图谱。采用Agilent ZORBAX SB-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-水为流动相进行梯度洗脱,流速1.0 ml/min,柱温30℃,检测波长203 nm。结果建立具有13个共有峰的栽培柴胡指纹图谱,各共有峰之间分离度较好,精密度、重复性和稳定性实验中各共有峰相对保留时间的RSD均小于1.0%,相对峰面积的RSD均小于5.0%。计算出各批次药材间的相似度。结论各批柴胡药材指纹图谱相似度大小与其裁培地区、采收期等因素相关,所建立的指纹图谱为柴胡的鉴别及质量控制提供了依据。

Abstract:Objective To establish chromatographic fingerprint of plant Bupleurum by HPLC which could be used to Bupleurum identity and quality controlling.Methods A method of HPLC-UV was applied to establish chromatographic fingerprint of 10 plant Bupleurum samples from different region and picking periods in Ningxia.An Agilent ZORBAX SB-C18(250 mm×4.6 mm,5 μm) column was used at 30 ℃,acetonitril-water was used as the mobile phase with gradient elution.The flow rate was 1.0 ml/min and the detection wavelength was 203 nm.Results The HPLC fingerprint of plant Bupleurum was set up with 13 common peaks and each peak in the fingerprint was well separated.The RSDs of the relative retention time of the common peaks were less than 1.0%.The RSDs of the relative peak area were less than 5.0%.The similarity of Radix Astragali from different region and picking periods was calculated.Conclusion The similarity of plant Bupleurum is related with its region and picking periods.This fingerprint is useful for the identity and quality controlling of plant Bupleurum
