摘 要: | 目的建立测定黄芩中黄芩苷、汉黄芩苷、黄芩素、汉黄芩素含量的HPLC方法,分析宁夏栽培黄芩中4种黄酮类成分的差异。方法采用迪马Platisil(铂金)ODS柱(250 mm×4.6 mm,5μm),以甲醇:乙腈:醋酸-醋酸钠(PH4.5)为流动相,梯度洗脱,检测波长278 nm,流速1.0 ml/min,柱温:40℃。结果黄芩苷、汉黄芩苷、黄芩素和汉黄芩素线性范围分别是0.07-15.43μg(r=0.999 8)、0.02-3.73μg(r=1.000 0)、0.007-1.44μg(r=0.999 9)、0.003-0.51μg(r=1.000 0),平均回收率分别为97.5%、98.7%、98.0%、95.5%;RSD分别为2.9%、1.9%、2.7%、1.3%。宁夏栽培黄芩样品中黄芩苷、黄芩素、汉黄芩苷和汉黄芩素的总含量之和范围在14.59%-23.29%。结论宁夏不同产地、不同生长年限、不同月份的黄芩样品中黄芩苷、黄芩素、汉黄芩苷和汉黄芩素的总含量存在一定差异,本法简便、准确、重复性好,可作为黄芩药材质量检测的方法。

Abstract: | Objective To establish a HPLC method for simultaneous determination of baica1in,wogonoside,baicalein and wogonin in cultivated Scutellariae and to study the difference of four flavonoids in cultivated scutellariae in Ningxia.Methods A Platisil ODS(250 mm×4.6 mm,5 μm) column was used at 40 ℃.Methanol:acetonitrile:acetate-sodium acetate(PH4.5) was used as the mobile phase with gradient elution.The detection wave length was 278 nm.The flow rate was 1.0 ml·min-1.Results The linear ranges of baicalin,wogonoside,baicalein and wogonin were respectively 0.07-15.43 μg(r =0.9998),0.02-3.73 μg(r =1.0000),0.007-1.44 μg(r =0.9999) and 0.003-0.51 μg(r =1.0000).The average recoveries were respectively 97.5%,98.7%,98.0% and 95.5%.And RSD were respectively 2.9%,1.9%,2.7% and 1.3%.The ranges of the content of baica1in,wogonoside,baicalein and wogonin in cultivated Scutellariae were 14.59%-23.29% in Ningxia.Conclusion The total content of baica1in,wogonoside,baicalein and wogonin in different months and different years and different localities in Ningxia have some difference.This method is simple,accurate and reliable.It can be used for the quality control of Scutellariae. |
