高血流肺动脉高压大鼠钙敏感受体、硫化氢及活性氧的研究

摘    要:目的:探讨肺动脉钙敏感受体(CaSR)、血浆硫化氢(H2S)、肺动脉内皮细胞中活性氧(ROS)水平在肺动脉高压发生发展中的作用。方法18只SD雄性大鼠按随机数字表法分为对照组和实验组,每组9只。实验组行左肺叶切除术,对照组行假手术,术后均喂养35 d。比较两组大鼠肺动脉压力( mPAP )、右心室肥厚指数、血浆H2 S、肺动脉内皮细胞ROS水平和肺动脉CaSR mRNA的表达水平。结果实验组大鼠mPAP、肺动脉内皮细胞ROS水平及右心室肥厚指数、肺动脉CaSR mRNA表达均明显高于对照组( P<0.05),血浆H2 S含量明显低于对照组( P<0.05)。结论肺高血流肺动脉高压大鼠肺动脉CaSR mRNA表达的改变和内皮细胞ROS对细胞的损伤可能通过改变内源性H2 S的产生而共同参与肺动脉高压的形成。

Abstract:Objective To explore the effect of pulmonary calcium sensitive receptor ( CaSR) ,plasma hydrogen sulfide ( H2 S) and reactive oxygen species ( ROS ) of pulmonary endothelial cells on the development of pulmonary arterial hypertension .Methods Eighteen male SD rats were randomly divided into control group and experimental group ,with nine rats in each group .The rats in the experimental group underwent left lung resection while in the control group underwent sham-operation ,and they were fed for 35 days after operation .The mean pulmonary arterial pressure (mPAP),right ventricle hypertrophy index (RVHI),plasma H2S,ROS level of pulmonary endothelial cells and mRNA expression of pulmonary CaSR were compared between two groups .Results The mPAP, ROS level of pulmonary endothelial cells,RVHI and mRNA expression of pulmonary CaSR of rats in the experimental group were significantly higher than those in the control group(P〈0.05),the plasma level of H2S of rats in the experimental group was signif-icantly lower than that in the control group ( P〈0 .05 ) .Conclusion The change of CaSR mRNA expression and the damage of ROS in endothelial cells in pulmonary hypertension rats with high blood flow may involve in the formation of pulmonary hypertension by changing the generation of endogenous H 2 S.
