摘    要:目的:建立测定刺薏胶囊中紫丁香苷含量的高效液相色谱法,并进行方法学考察。方法:采用梯度洗脱法,用ZORBAXSB-C18(5μm,4.6mm×150mm)色谱柱,以甲醇为流动相A,水为流动相B,以1.0ml/min的流速进行梯度洗脱(0~20min,A为18%;20~30min,A为18%~70%;30~35min,A从70%~18%);检测波长265nm。结果:紫丁香苷进样量在0.0808~0.404μg与峰面积呈良好的线性关系,r=0.9999;平均回收率为98.2%,RSD为1.75%。结论:梯度洗脱反相HPLC法测定刺薏胶囊中紫丁香苷含量的方法专属性强、准确稳定、重复性好,可用于刺薏胶囊的质量监控。

Abstract:Objective: To establish a method for the determination of syringoside in Ciyi compound preparation with reverse phase high pressure liquid chromatography (RP-HPLC) and to evaluate the methodology. Methods: The column was ZORBAX SB-C18(5μm,4.6mm×150mm ). The flow rate was 1.0 ml/min. The mobile phase A was methanol and mobile phase B water. The procedures were that: at ranges of 0 -20 min, 20 -30, and 30 -35 min, concentrations of A were 18% , 18% - 70%, and 70% - 18% respectively. The detection wavelength was 265 nm. Results: When the sample sizes were in the range of 0. 080 8 - 0. 404 μg, the standard curve for syringoside was satisfactorily linear ( r = 0. 999 9). The average recovery was 98.2% with RSD = 1.75% ( n = 6). Conclusions: The method is specific, accurate, and stable, and it can be used for detemination of syringoside in Ciyi compound preparation.
