摘    要:①目的探讨HIV-1是否能够侵染及整合在多种组织细胞的DNA中。②方法采用套式PCR对来自艾滋病病人的肺、脑、肾、肌肉及口腔脱落细胞标本进行HIV-1前病毒pol基因的检测,并根据外侧引物PCR和套式引物PCR敏感性对原始模板数量进行半定量检测。③结果采用套式引物PCR可以在艾滋病病人的肺、脑、肾、肌肉及口腔脱落细胞DNA标本中检出HIV-1前病毒pol基因,而采用外侧引物PCR仅在肺、脑细胞DNA标本中检出了该基因。④结论HIV-1可以侵染机体的多种组织器官并发生整合

Abstract:Objective To confirm whether HIV 1 could infect and integrate into cellular DNA of multiple tissues. Methods The nested primers polymerase chain reaction technique was employed to detect the proviral genome of HIV 1 pol gene integrated in cellular DNA of pathologial tissues including lung, brain,liver, kidney, muscle and cast off cells of oral mucosa from a dead AIDS patient. The number of primary template of HIV 1 pol gene integrated in the cellular DNA of multiple tissues was postulated by half quantitative analysis with the routine and nested PCR. Results The proviral genome of HIV 1 pol gene could be detected in cellular DNA of all pathological tissues by nested PCR, while the gene could only be detected in lung, brain and liver tissues by routine PCR. Conclusion The HIV 1 could infect and integrate into multiple tissues without obviously pathological changes.
