摘    要:目的探讨非小细胞肺癌(NSCLC)患者呼出气冷凝液(EBC)中p53基因突变检测的临床意义。方法采用巢式PCR结合DNA测序法,检测53例NSCLC患者EBC中p53基因第5~8外显子的突变情况,并以32例健康体检者EBC标本作为对照。结果肺癌组53例EBC标本中扩增到1)53基因26例,其中10例检测到p53基因突变,突变率为38.5%(10/26);正常对照组32例EBC标本中扩增到p53基因15例,但未检测到p53基因突变。结论p53基因突变可能参与NSCLC的发生发展,检测EBC中p53基因的突变对肺癌的早期诊断有一定的价值。

Abstract:Objective To investigate the clinical value ofdetectingp53 gene mutation in EBC from patients with non-small cell lung cancer (NSCLC). Methods Polymerase chain reaction and DNA sequencing method were performed to examine the point mutations in the conserved exon 5,6,7 and 8 of p53 gene in EBC from 53 cases of patients with NSCLC and matched 32 healthy control. Results The positive amplification ofp53 gene were detected in 26 from 53 cases of patients with NSCLC, while in 15 from 32 healthy control. The mutation of p53 gene in patients with NSCLC was 38.5%(10/26), while no mutation were found in healthy control. Conclusion Thep53 gene mutation may be involved in the occurrence and development of NSCLC and the monitoringp53 gene mutation may be helpful to the early diagnosis of lung cancer.
