川芎嗪防治特发性肺纤维化的MMP表达

摘    要:目的探讨川芎嗪对博来霉素诱导IPF模型的干预作用及相关机制。方法 80只SD大鼠随机分为4组:假手术组、模型组、阳性药物对照组和川芎嗪组。除假手术组外其余各组气管灌注博来霉素诱导肺纤维化,假手术组灌注生理盐水。术后第2天开始灌胃给药,阳性药物对照组灌胃醋酸强的松1.5mg.kg-1.d-1,川芎嗪组给予川芎嗪20mg.kg-1.d-1,模型组和假手术组给予相应体积的生理盐水。分别于第14天和第28天每组各取10只处死,取肺组织,HE染色和Mallory染色观察肺泡炎和纤维化程度,免疫组化法检测MMP-1的表达。结果川芎嗪可改善IPF模型大鼠肺泡炎和肺纤维化程度。川芎嗪组MMP-1表达较模型组降低。结论川芎嗪可减轻博来霉素诱导的IPF大鼠肺纤维化程度,减少MMP-1表达,可能是其抗IPF的机制之一。

Abstract:Objective To investigate the influence of TMP on bleomycin-induced pulmonary fibrosis in rats and explore its mechanism.Methods 80 SD rats were randomly divided into four groups: sham group,model group,prednisone group and TMP group.The rats were received injection with a single dose of bleomycin by trachea except sham group,and rats in sham group with same volume normal saline.One day after the injection,prednisone(1.5mg·kg-1·d-1) and TMP(20mg·kg-1·d-1) were respectively given to those in the prednisone group and TMP group daily by gastrogavage,while equal volume of saline was given to those in sham group and model group.On the 14th,28th day,10 rats per group were sacrificed and the lung was incised to make pathological sections which were stained with HE and Mallory.The expression of MMP-1 in lung was determined with immunohistochemisty.Results TMP could inhibit the airsacculitis and pulmonary fibrosis and reduce the expression of MMP-1 in lung tissue.Conclusion TMP can reduce the extent of alveolitis and fibrosis in bleomycin-induced pulmonary fibrosis and repress the expression of MMP-1,which may be relevant to the mechanism of its protective effects on IPF.
