p53基因和血管生成抑制素基因共转染对胃癌SGC7901细胞凋亡的影响

摘    要:目的:探讨血管生成抑制素(angiostatin,AS)基因和p53基因共转染人胃癌细胞株SG7901后对其凋亡的影响. 方法:分别将p53、AS基因以及p53联合AS基因转染SG7901细胞;采用RT-PCR法检测转染后SG7901细胞中目的基因的表达;通过细胞集落形成实验和MTT法观察不同转染对细胞的生长抑制作用;FCM法检测p53基因和AS基因对SGC7901凋亡的影响.结果:经p53或AS基因转染后,SG7901细胞的集落数量及大小均有不同程度的降低;且p53和AS基因具有协同作用,以共转染组降低最为明显,差异有统计学意义(P<0.05).MTT法检测结果表明,共转染组细胞的生长情况较其他2组单基因转染的细胞更为缓慢(P<0.05).共转染组凋亡率较AS基因单转染组、p53基因单转染组以及空载体转染组均高,差异有统计学意义(P<0.05).结论:p53和AS基因均能诱导胃癌细胞SG7901的凋亡,且两者具有协同作用.
