| 摘 要： | 目的:观察中药止喘汤对哮喘大鼠气道重构的干预,并探讨其对基质金属蛋白酶-9(MMP-9)及金属蛋白酶抑制剂-1(TIMP-1)表达的影响。方法:50只Sprague-Dawley(SD)大鼠随机分为正常组、哮喘组、布地奈德(BUD)组、黄芩组及止喘汤组5组。采用卵清白蛋白(OVA)致敏加激发法复制大鼠哮喘模型,苏木精-伊红染色测气道形态学参数,免疫组化染色测定气道壁MMP-9、TIMP-1及IV型胶原含量。结果:哮喘组较正常组大鼠气道壁明显增厚,MMP-9、TIMP-1表达增多(P<0.01)。用中药和BUD干预后大鼠气道壁明显变薄,MMP-9、TIMP-1表达减少,与哮喘组比较有统计学意义(P<0.01);其中复方组肺纤维化轻于单味组,各项指标比较有统计学意义(P<0.05)。大鼠气道壁的厚度、IV型胶原含量、MMP-9、TIMP-1及MMP-9/TIMP间有相关关系。结论:中药具有干预哮喘大鼠模型气道重构的作用,这些作用可能是通过调节MMP-9和TIMP-1而实现的,基于中医基本病机证型的复方优于单味中药。
