母体暴露壬基酚对仔鼠脾脏组织的氧化损伤

摘    要:目的]研究母体暴露壬基酚(NP)对仔鼠脾脏的氧化损伤。方法]将SD母鼠随机分为50、100、200mg/kgNP暴露组及对照组,每组6~7只动物。在母鼠妊娠第9~15天灌胃NP,仔鼠(F1)于出生印天龄时处死并取脾脏称重。比色法测定脾脏组织中超氧化物歧化酶(SOD)活力及脂质过氧化产物丙二醛(MDA)含量;透射电镜观察其超微结构改变。结果]与对照组比较,200mg/kgNP组仔鼠的脾脏重量及脏器系数均明显降低(P〈0.05),且与染毒剂量呈负相关(r脾=-0.618,r脏=-0.530,P〈0.05);100、200mg/kgNP组仔鼠脾脏组织的SOD活力降低,200mg/kgNP组MDA含量升高(P〈0.05),均存在剂量效应关系(rSOD=-0.460,rMnA=0.747,P〈0.05)。高暴露组仔鼠脾脏电镜下可见脾细胞线粒体肿胀呈空泡样变,染色质浓缩成块状聚集在核周围。结论]NP可透过胎盘屏障致仔鼠脾脏脂质过氧化损伤。

Abstract: Objective ] To investigate the adverse effects on F] rats'spleen while the pregnant mother rats were exposed to nonylphenol ( NP ). Methods ] Pregnant rats were exposed to NP via gavage at doses of 0, 50, 100 and 200 mg/kg by body weights respectively per day, from pregnant days 9 to 15. The level of malondialdehyde( MDA )and activity of super oxide dimutese ( SOD ) in FI rats' spleen homogenate were detected respectively at the age of 60th day. The ultrastructure of spleen tissues were observed under electronic microscope. Results ] Compared with the control group, the absolute and relative weight of spleen were decreased dramatically in the 200 mg/kg NP treated group and negative relation between the weight and dose was found ( rSOD=-0.618, rcoefficient=-0.530, P〈0.05 ). The SOD activity of spleen tissues decreased in NP I00 and 200 mg/kg exposed fetal rats, while the levels of MDA increased in NP 200mg/kg exposed fetal rats. Both of them were in a dose-dependent manner (rSOD=-0.460, rMDA=0.747, P〈0.05 ). The differences mentioned above were statistical significant compared to control group( P〈0.05 ). The changes of the uhrastructrue were found among the spleen of NP 200 mg/kg group that characterized with ehromatin condesed, clumped in circa-nuclei and mitochondrial tumefaction and vaeuolization. Conclusion ] NP can get across the placenta barrier to make the F1 offspring rats'spleen damage of lipid peroxidation.
