摘    要:目的探讨施万细胞(SCs)在蚕丝支架上体外三维培养的生长活性。方法将所培养的乳鼠面神经SCs与蚕丝支架复合培养,用倒置显微镜、扫描电镜、透射电镜和四甲基偶氮唑蓝(MTT)法观察并测定SCs在蚕丝支架上生长形态及活性。结果蚕丝支架上SCs的形态及活性指标与正常培养的SCs比较差异无显著性(P>0.05)。结论蚕丝支架能维持SCs的正常形态和活性,蚕丝是SCs立体培养时良好的天然支架。

Abstract:Objective To detect the activity of Schwann cells(SCs) cultured three-dimensionally on silk scaffold in vitro.Methods Cultured SCs of facial nerve in neonatal rats were seeded in silk scaffold,and then we observed and measured the shape and activity of SCs by inverted microscopy,scanning electronic microscope,transmission electronic microscope and MTT method.Results There was no difference between the activity of SCs cultured on silk scaffold and those of control cells(P>0.05),and no difference in cellar shape was observed. Conclusion Silk scaffolds are ideal for shape and activity maintenance of SCs,and silks can be used as natural scaffolds for SCs in three-dimensional culture.
