银杏叶提取物对糖尿病大鼠视网膜神经节细胞Bax和Bcl-2基因表达的影响

摘    要:目的 观察银杏叶提取物(extract of ginkgo biloba leaf, EGB)对糖尿病大鼠视网膜神经节细胞凋亡相关基因Bax和Bcl-2表达的影响,探讨EGB防治糖尿病性视网膜病变的作用机制.方法 选择健康成年Wistar大鼠30 只, 随机分成正常对照组、模型组和EGB治疗组.腹腔内注射链脲佐菌素(STZ) 诱发大鼠糖尿病.EGB治疗组给予EGB溶液灌胃12周,模型组和对照组灌服等体积生理盐水,随后制备视网膜石蜡切片,行免疫组化染色, 并对染色结果行计算机图像分析.透射电镜观察各组神经节细胞的超微结构变化.结果 Bax在模型组神经节细胞的表达较正常对照组显著增强,在EGB治疗组的表达强度较模型组减弱;Bcl-2在EGB治疗组表达较模型组明显增强,差异均有统计学意义(P<0.01).EGB治疗组神经节细胞超微结构的损害明显减轻.结论 EGB可减轻糖尿病大鼠视网膜神经节细胞的超微结构损伤,减轻细胞凋亡的发生,其机制可能与EGB调节凋亡相关基因Bax和Bcl-2的表达有关.
