摘    要:目的:研究糖皮质激素对Graves病(GD)患者甲状腺细胞凋亡的影响及其临床意义。方法:以不同浓度地塞米松刺激单层培养的GD甲状腺上皮细胞(TEC),采用流式细胞术,ELISA法及半定量RT-PCR法检测刺激前后细胞凋亡率,Fas表达阳性率,可溶性Fas(sFas)含量以及Fas/sFasmRNA表达的变化。结果:(1)在0-10^3nM范围内,地塞米松可剂量依赖性地促进GD TEC凋亡;(2)在上述浓度区间,地塞米松可显著增加GD TEC Fas蛋白及mRNA的表达,抑制sFas蛋白及mRNA的产生。(3)Fas蛋白表达增加,aFas蛋白分泌减少与细胞凋亡间存在显著相关性。结论:糖皮质激素可显著促进甲状腺细胞凋亡的发生,促进Fas合成,抑制sFas产生,导致甲状腺功能异常。

Abstract:Objective To investigate the effect of glucocorticoids on apoptosis in thyrocytes from patients with Graves disease(GD).Methods Thyrocytes from GD were cultured in the absence or presence of 10~103nM dexamethasone.Apoptosis,Fas expression,soluble Fas(sFas) and Fas/sFas mRNA level in thyrocytes were detected by flow cytometry,ELISA and semi-quantitative RT-PCR.Results (1)Dexamethasone in 10~103nM could increase apoptosis dose-dependently(P<0.01).(2)Fas expression and Fas mRNA level in GD thyrocytes were markedly increased by dexamethasone,whereas sFas and sFas mRNA level were significantly decreased by dexamethasone(P<0.05).(3)The rate of apoptosis was positively correlated with Fas expression(r=0.96,P<0.01),but negatively correlated with sFas level(r2=-0.95,P<0.05).Conclusion Dexamethasone could induce apoptosis in GD thyrocytes in vitro through increased expression of Fas and decreased production of sFas.
