超声对大鼠坐骨神经损伤后神经生长因子蛋白表达的影响

摘    要:目的研究低强度超声对大鼠坐骨神经损伤后神经生长因子(NGF)蛋白表达的影响。方法以64只成年雌性W istar大白鼠右侧坐骨神经重度钳夹伤为动物模型,应用随机数字表进行随机化,将实验动物均分为超声组与对照组。按取材时间分为术后2、4、6、8周等4个时间段(每组8只)。超声组的造模侧神经损伤处给予超声辐照,对照组相应部位予以未启动治疗系统的假辐照。术后不同时间取紧邻挤压伤部位的远端坐骨神经干,应用免疫组织化学和图像分析的方法研究神经组织中NGF的表达并进行定量分析。结果术后4、6、8周,超声组坐骨神经组织中的NGF蛋白高表达。结论低强度超声能够促进大鼠坐骨神经损伤后神经组织NGF蛋白的表达。

Effects of low-intensity ultrasound on the expression of nerve growth factor after sciatic nerve injury in rats

Abstract:Objective To study the effects of low-intensity ultrasound(LIU) on the expression of nerve growth factor(NGF) after sciatic nerve injury in rats.Methods The right sciatic nerves of 64 adult Wistar rats were crushed with forceps for the experimental animal models and they were randomly divided into treatment group and control group.The rats in the treatment group received the LIU exposure,and rats in the control group received with sham exposure.The rats in the two groups were equally subdivided into 4 groups according to the different postoperative interval: 2,4,6,and 8 weeks.At various intervals after surgery,the sciatic nerve stump immediately distal to the crushed site was harvested.The expression of NGF was evaluated by using Immunohistochemistry and image analysis.Results The expression of NGF in the sciatic nerve tissue was higher in LIU group than that of the control group at the fourth,sixth and eighth weeks after surgery.Conclusion LIU can promote the expression of NGF protein after sciatic nerve injuries in rats.
