环孢菌素A对兔视网膜色素上皮细胞光损伤防治作用的观察

摘    要:目的研究环孢菌素A(cyclosporine A)对兔视网膜色素上皮光损伤的防治应用。方法取兔眼的视网膜色素上皮细胞作培养。分为4组,一组阳性对照,一组阴性对照,另外两组分别于不同时间给予环孢菌素A。光照后,应用吖啶橙染色观察各个组别的细胞凋亡情况。结果光照后,阳性对照组出现明显的凋亡,细胞吖啶橙染色出现凋亡的形态学改变。光照前加药组凋亡不明显。而后加药组则出现明显凋亡,和阳性对照组相比并无差别。结论环孢菌素A可以预防视网膜色素上皮细胞光损伤所致的凋亡,其机制可能是与细胞线粒体相结合而预防凋亡。

Abstract:Objective To study the effect of prevention and therapy of cyclosporine A on retinal pigment epithelium cell of rabbits. Methods The Retinal pigment epithelium ceils of rabbits were cultured by dividing them into 4 groups. One group was used as positive control, the other was used as negative control, others were treated by cyclosporine A at different times. They were exposed to the light for an hour, and then the apoptosis of each group was observed by acridine orange. Results The positive control exhibited obvious apoptosis when treated by light, apoptotic morphological changes were observed by fluorescence microscope. The apoptosis in the group which was given eyelosporin A before photic inju- ry was not obvious. However, it was apparent in the group which was given eyelosporin A after photic injury. It was not different compared with the positive control. Conclusions Cyclosporin A could prevent the retinal pigment epithelium from apoptosis caused by light. The prevention of apoptosis is probably associated with the combination between cyclosporin A and mitochondrion in cell.
