益胃冲剂和米索前列醇对大鼠胃粘膜PGE2及血流量的影响

目的 :观察“益胃冲剂”和米索前列醇对大鼠胃粘膜内源性PGE2 及血流量的影响。方法 :采用吲哚美辛为胃粘膜损伤剂 ,米索前列醇为对照药物 ,Ⅱ级雄性SD大鼠 ;完全随机化平均分为 :控制组、损伤组、益胃冲剂治疗组和米索前列醇治疗组 ;分别放免检测胃粘膜PGE2 含量 ;激光多普勒血流仪检测胃粘膜血流量和病理切片镜下计算胃粘膜病理损伤长度。结果 :益胃冲剂和米索前列醇均可保持胃粘膜PGE2 含量和GMPF明显高于吲哚美辛损伤组 (P <0 .0 1 ) ,但也明显低于控制组 (P <0 .0 1 )的水平 ;胃体部PGE2 含量米索前列醇优于益胃冲剂 (P <0 .0 1 ) ,胃窦部却无差别 ;治疗组几乎不出现Ⅲ级病理损伤。结论 :益胃冲剂和米索前列醇均呈现良好的胃粘膜保护作用

Effects of Yiwei extract on the gastric mucosal PGE2 and gastric mucosal blood flow in rats

Objective: To observe the effect of Yiwei extract supplementing Qi and nourishing Yin on local defensive mechanism of gastric mucosa. Methods:Sixty four SD rats weighing 180-220 g were randomly classified into four groups: control group (S), Indomethacin injury group (M),Yiwei extract treatment group (YM), Misoprostol treatment group (PM). Indomethacin, a gastric mucosa injury agent, was injected into rat stomach. Saline solution, saline solution plus indomethacin, Yiwei extract plus Indomethacin and Misoprotol plus Indomethacin were given to group S, group M, group YM and group PM, respectively. Four hours after the last administration, 8 rats of each group were killed euthanasically to get gastric mucosa for the measurement of endogenous prostaglandin E 2(PGE 2) concentration of gastric mucosa lysate by radioimmunoassay; the left 8 rats in each group were anesthetized and gastric mucosal blood were measured by Doppler blood flow meter followed by pathological examination.Results:The content of endogenous PGE 2 in mucosa of gastric corpus was significantly higher ( P <0.01) in group YM compared with group M, and so did it in gastric antrum .Gastric mucosal blood flow was higher in group YM and group PM compared with those in group S and group M.Conclusion: Yiwei extract is able to effectively antagonize the Idomethacin-induced injury of gastric mucosa by means of promoting the synthesis of endogenous PGE 2 and enhancing the gastric mucosal blood flow.
