摘 要: | 目的评价罗格列酮预先给药对2型糖尿病大鼠肾缺血再灌注损伤(I/R)的影响。方法健康清洁级成年雄性SD大鼠,体重180-220g,采用高脂高糖饲料喂养加用小剂量链脲佐菌素注射的方法制备2型糖尿病大鼠模型。取糖尿病模型制备成功的大鼠30只,采用随机数字表法,将其随机分为3组(n=10):假手术组(S组)、缺血再灌注组(I/R组)和罗格列酮组(R组)。于I/R模型建立前给药,R组给予罗格列酮3mg/(kg·d)灌胃,S组和I/R组给予等容量生理盐水,连续10d。给药结束后I/R组和R组采用切除右肾,夹闭左肾动、静脉45min后恢复灌注的方法制备大鼠肾缺血再灌注模型,S组切除右肾,不夹闭左肾动、静脉。分别于再灌注24h时抽取腹主动脉血,测定血清肌酐(Cr)和尿素氮(BUN)浓度,然后处死大鼠取肾,光镜下观察肾组织形态学变化并进行组织学评分,免疫组化法测定NF-κB表达。结果与S组比较,I/R组和R组血清肌酐(Cr)及尿素氮(BUN)浓度、肾组织学评分及NF-κB表达升高(P〈0.05);与I/R组比较,R组血清肌酐(Cr)及尿素氮(BUN)浓度、肾组织学评分及NF-κB表达降低(P〈0.05)。结论罗格列酮可减轻糖尿病大鼠肾缺血再灌注损伤,其机制可能与抑制NF-κB表达有关。

Abstract: | Objective To evaluate the effects of rosiglitazone pretreatment of type 2 diabetic rats with renal ischemia-reperfusion injury(I/R). Methods Healthy cleaned adult male SD rats weighing 180-220g,high-fat high-carbohydrate diet with a small dose of streptozotocin injection method in type 2 diabetic rat model was prepared. A successful preparation of diabetic rat model 30 was taked,using a random number table,randomly divided into 3 groups(n=10):sham operation group(S group),ischemia-reperfusion group(I/R group) and rosiglitazone group(R group). Established prior to administration in I/R model,R group received rosiglitazone 3mg/(kg·d) orally,S group and I/R group received the same volume of saline,continuous 10d. After the administration of I/R group and the R group with bilateral renal artery occlusion,45min after intravenous infusion recovery process for the preparation of rat renal ischemia-reperfusion model,S group does not clamping bilateral renal artery and vein. We drawn at 24h reperfusion abdominal aortic blood,serum creatinine(Cr) and blood urea nitrogen(BUN) concentration,and then the rats were sacrificed kidney,light microscopy and histological changes in renal histological scores,immunohistochemistry determination of the method NF-κB expression. Results Compared with S group,I/R group and the R group,serum creatinine(Cr) and blood urea nitrogen(BUN) concentration and kidney histology scores and NF-κB expression was increased(P〈0.05),compared with the I/R group, R serum creatinine(Cr) and blood urea nitrogen(BUN) concentration and kidney histology scores and NF-κB expression decreased(P〈0.05). Conclusion Rosiglitazone can reduce renal ischemia-reperfusion injury in diabetic rats,which may be related to inhibition of NF-κB expression. |
