雀脑药材的质量标准提升研究

摘    要:目的 进行雀脑药材的鉴别、检查及含量测定研究,为其临床应用、生产使用与市场监督管理提供质量保证。方法 鉴别研究采用显微鉴别及薄层鉴别方法;水分检查采用烘干法,总灰分及酸不溶性灰分检查采用炽灼残渣法;含量测定采用总氮量法及全自动氨基酸分析仪法。结果 雀脑的显微鉴别特征为树突碎片与神经纤维;薄层鉴别以丙氨酸为对照,鉴别氨基酸类成分,随机选取20111001批雀脑药材为对照药材。水分介于5.57%-5.83%,总灰分介于4.40%-4.70%,酸不溶性总灰分介于0.28%-0.38%。总氮量介于6.49%-7.04%。17种氨基酸的线性相关系数介于0.9997-0.9999;精密度实验RSD介于0.11%-0.52%;10小时内稳定性良好,RSD介于0.02%-0.84%;重复性实验RSD介于0.17%-2.27%;回收率实验RSD介于1.24%-2.47%,平均回收率介于97.38%-102.19%;3批雀脑药材的17种氨基酸总含量介于47.61-66.37 mg·g-1结论 建立显微鉴别、薄层鉴别及含量测定方法专属性强,重现性好,具有较强创新性,对于控制雀脑药材质量具有显著意义;水分、总灰分、及酸不溶性灰分测定结果,可以为制订雀脑药材质量标准提供实验依据。

Abstract:Objective To study the identification, the inspection and the content determination of sparrow-brains, in order to provide the quality assurance for its use of clinical or production, and for supervision and management of market.Methods The microscopic identification and TLC were used for identification, the drying method was used for inspection of moisture, and the burning residue ethod was used for inspection of total ash and acid-insoluble ash. The total nitrogen method and automatic analyzer were used for content determination.Results The microscopic characteristics were dendrites and nerve fibers. TLC identification were used for alanine as reference substance to identify amino acid components, and to randomLy select 20111001 batche sparrow-brains as control species. The water content ranged 5.57%-5.83%, the total ash content ranged 4.40%-4.70%, and the acid insoluble ash content ranged 0.28%-0.38%. The total nitrogen ranged 6.49%-7.04%. The linear correlation coefficient of 17 amino acids ranged 0.9997-0.9999. RSD of precision experiment ranged 0.11%-0.52%. Stability within 10 hours was good, RSD ranged 0.02%-0.84%. RSD of repetitive experiment ranged 0.17%-2.27%. RSD of recovery experiment ranged 1.24%-2.47%, average recovery rate ranged 97.38%-102.19%. The total content of 17 amino acids of 3 batches of sparrow-brains ranged 47.61 -66.37 mg/g.Conclusion The establishment of microscopic identification, TLC identification and content determination methods is specific, reproducible and innovative, which has significance to controll the quality of sparrow brains. The results of moisture, total ash and acid-insoluble ash detection can provide experimental basis for the quality standards of sparrow-brains.
