摘    要:目的研究回转模拟微重力效应对Wistar大鼠皮层神经元蛋白质羰基化的影响。方法利用回转器模拟微重力 ,将原代培养的Wistar大鼠皮层神经元分别回转 0h、1 2h、2 4h、36h、48h。提取细胞总蛋白 ,酶联免疫吸附法 (EILSA)检测羰基化蛋白的水平。结果与 1G对照组相比 ,回转 2 4h、36h、48h显著增加皮层神经元羰基化蛋白的水平 (P <0 .0 5 )。结论回转显著增加皮层神经元羰基化蛋白的水平 ,这可能是神经元损伤的重要原因之一。

Abstract:Objective To investigate the effects of simulated microgravity on carbonyl protein in primary culture of neuron from Wistar rats. Method Primary culture of neurons from Wistar rats were exposed to simulated microgravity on a clinostat for 0 h,12 h,24 h,36 h,48 h. Total content of carbonyl protein in cell was isolated and determined by enzyme-linked immunosorbent assay and time courses of protein oxidation following simulated microgravity were observed. Result Protein carbonyl significantly increased after 24 h,36 h and 48 h(P<0.05) of clinostat as compared with 1G control group. Conclusion The increase of carbonyl protein content caused by simulated weightlessness is likely to be one of the important reasons of neuron injury.
