摘    要:目的:通过测定食管鳞状细胞癌(食管鳞癌)患者肿瘤组织中程序性死亡4基因(Pdcd4)的表达,探讨Pdcd4与食管鳞癌发生、发展的关系。方法:应用实时荧光定量聚合酶链反应检测60例食管鳞癌组织中Pdcd4 mRNA的表达情况,同时应用免疫组织化学法检测石蜡包埋的178例食管鳞癌组织中Pdcd4的表达水平,相应的癌旁正常组织作为对照。结果:Pdcd4 mRNA在食管鳞癌组织中的表达量显著低于癌旁正常组织(P<0.01)。Pdcd4在癌旁正常组织中的阳性表达率为100.0%,明显高于食管鳞癌组织中的47.2%(P<0.01)。Pdcd4的表达与肿瘤的分化程度有关,在高分化食管鳞癌的阳性率为71.4%,中分化食管鳞癌的阳性率为43.4%,低分化食管鳞癌的阳性率为30.2%,高分化鳞癌中Pdcd4阳性率高于中分化及低分化癌(P<0.01);中、低分化食管鳞癌中Pdcd4阳性率差异均无统计学意义(P >0.05);Pdcd4阳性率在患者的年龄、肿瘤大小、TNM分期和N分期间差异均无统计学意义(P >0.05)。结论:Pdcd4异常表达与食管鳞癌的组织病理分级有关。

Abstract:Objective:To explore the expression of programmed cell death 4(Pdcd4) gene in esophageal squamous cell carcinoma, and relationship between the expression level of Pdcd4 and occurrence and development of tumor. Methods:The expressions of Pdcd4 mRNA in 60 esophageal squamous cell carcinoma tissues and adjacent normal tissue were detected by real-time polymerase chain reaction,and the expressions of Pdcd4 protein in 178 esophageal squamous cell carcinoma tissues and adjacent normal tissue were detected by immunohistochemical method. Results:The expression of Pdcd4 mRNA in esophageal squamous cell carcinoma tissue were significantly lower than that in adjacent normal tissue(P<0. 01). The positive expression rate of Pdcd4 in esophageal squamous cell carcinoma tissue(47. 2%) were significantly lower than that in adjacent normal tissue(100. 0%)(P<0. 01). The expression of Pdcd4 was correlation with tumor differentiation, the positive expression rates of Pdcd4 in high, moderate and low differentiation esophageal squamous cell carcinoma tissue were 71. 4%, 43. 4% and 30. 2%, respectively. The positive expression rate of Pdcd4 in high differentiation esophageal squamous cell carcinoma tissue was higher than that in moderate and low differentiation esophageal squamous cell carcinoma tissue, the difference of the positive expression rate of Pdcd4 between moderate and low differentiation esophageal squamous cell carcinoma was not statistically significant(P>0. 05). The difference of the positive expression rate of Pdcd4 between age of patient,tumor size,TNM staging and N staging were not statistically significant(P>0. 05). Conclusions:The abnormal expression of Pdcd4 is correlation with the occurrence and development of esophageal squamous cell carcinoma.
